Could be an end knock. From experience I found the knocking sound tends to vary with engine load. But others might know better!!

Possible a worn valve gear or camshaft
Is it doing it when cold then it could be piston slap?
or it could be component fault ie coolent pump

It could be the big end bearing less noise when engine is under load
or main bearing more noise under load,

I wouldn't have thought valve gear noise would be dependant upon load. But then one never knows. As for piston slap, again I would've thought more noticeable on load. Seem to remember in the long and distant past (very distant!), a scooter I had made a 'orrible noise when rolling back the throttle. Discovered that the small end bearing was knackered, such the the gudgeon pin hole in the piston was more slot like!!!!
